Potential genomic biomarkers of obesity and its comorbidities for phthalates and bisphenol A mixture: In silico toxicogenomic approach

This in silico toxicogenomic study aims to explore the relationship between phthalates and bisphenol A (BPA) co-exposure obesity, as well its comorbid conditions, order construct a possible set of genomic biomarkers. The Comparative Toxicogenomics Database (CTD; http://ctd.mdibl.org) was used main data mining tool, along with GeneMania (https://genemania.org), ToppGene Suite (https://toppgene.cchmc.org) DisGeNET (http://www.disgenet.org). Among phthalates, bis(2-ethylhexyl) phthalate (DEHP) dibutyl (DBP) were chosen most frequently curated CTD, which also share similar mechanisms toxicity. DEHP, DBP BPA interacted 84, 90 194 obesity-related genes/proteins, involved 67, 65 116 pathways, respectively. these, 53 genes/proteins 42 pathways common all three substances. 31 had matching interactions for investigated substances, while more than half these (56.49%) co-expression. 7 (6 relevant humans: CCL2, IL6, LPL, PPARG, SERPINE1, TNF) identified obesity comorbidities, PPARG LPL closely linked obesity. These could serve target further vitro vivo studies molecular mixture obesogenic properties. Analysis reported here should be applicable any environmental chemicals disease present CTD.
